Abstract

ABSTRACT

Blood glucose levels may change while specimens are awaiting analysis, particularly when cellular components remain in the specimen and storage conditions vary. This issue warrants attention because changes during the pre-analytical stage may affect the interpretation of glucose test results. This study evaluated changes in fasting blood glucose levels following an 8-hour delay in analysis by considering serum separation and storage temperature. A total of 25 fasting blood specimens were examined using the Glucose Oxidase Phenol Aminophenazone (GOD-PAP) method with a Mindray BS-200 Chemistry Analyzer. Each specimen was assessed under five conditions: immediate analysis, separated serum stored at room temperature, separated serum stored in a refrigerator, unseparated specimens stored at room temperature, and unseparated specimens stored in a refrigerator. The respective mean glucose levels were 179.80, 176.60, 175.40, 159.00, and 171.20 mg/dL. Kruskal-Wallis analysis yielded p=0.691 (p>0.05), indicating no statistically significant difference in glucose levels among the observed conditions. Descriptively, the greatest change occurred in specimens that remained in contact with blood cells during storage at room temperature. These findings highlight the importance of controlling specimen conditions when analysis is delayed and provide a basis for further studies involving larger sample sizes and broader storage durations.

ABSTRAK

Stabilitas kadar glukosa darah dapat berubah selama spesimen menunggu pemeriksaan, terutama ketika komponen seluler masih berada dalam spesimen dan kondisi penyimpanannya berbeda. Persoalan ini penting diperhatikan karena perubahan pada tahap pre-analytical dapat memengaruhi interpretasi hasil pemeriksaan glukosa. Penelitian ini mengevaluasi perubahan kadar glukosa darah puasa setelah penundaan pemeriksaan selama 8 jam dengan mempertimbangkan pemisahan serum dan suhu penyimpanan. Sebanyak 25 spesimen darah puasa diperiksa menggunakan metode Glucose Oxidase Phenol Aminophenazone (GOD-PAP) pada Mindray BS-200 Chemistry Analyzer. Setiap spesimen diamati pada lima kondisi, yaitu pemeriksaan segera, serum dipisahkan pada suhu ruang, serum dipisahkan pada suhu refrigerator, spesimen tidak dipisahkan dari sel darah pada suhu ruang, dan spesimen tidak dipisahkan pada suhu refrigerator. Nilai rerata berturut-turut sebesar 179,80; 176,60; 175,40; 159,00; dan 171,20 mg/dL. Analisis Kruskal-Wallis menghasilkan p=0,691 (p>0,05), sehingga belum ditemukan perbedaan kadar glukosa yang bermakna secara statistik antar kondisi yang diamati. Secara deskriptif, perubahan terbesar tampak pada spesimen yang tetap bersama sel darah selama penyimpanan suhu ruang. Temuan ini menegaskan pentingnya pengendalian kondisi spesimen selama penundaan pemeriksaan dan menjadi dasar untuk penelitian lanjutan dengan jumlah spesimen lebih besar serta variasi waktu penyimpanan yang lebih luas.
