Abstract

ABSTRACT

Staphylococcus aureus is a pathogenic bacterium that can be found in diabetic ulcers and may impede the wound-healing process. Inappropriate use of antibiotics can increase the risk of bacterial resistance, highlighting the need to explore natural substances as alternative antibacterial agents. This study aimed to analyze the antibacterial activity of garlic infusion (Allium sativum) against clinical isolates of S. aureus from diabetic ulcers based on bacterial colony counts. The infusion was prepared at concentrations of 50%, 65%, 80%, and 95%, with four replicates for each treatment group. Antibacterial activity was evaluated using a broth dilution method, followed by inoculation onto solid media and colony counting. Data were analyzed using the Shapiro–Wilk test, Levene’s test, and One-Way ANOVA at a 5% significance level. The results showed a significant difference in bacterial colony counts among the concentration groups (p < 0.05), with colony counts decreasing as the infusion concentration increased and the highest inhibitory activity observed at the 95% concentration. These findings indicate that garlic infusion exhibits antibacterial activity against clinical isolates of S. aureus from diabetic ulcers and shows potential as a natural-based antibacterial candidate, particularly at a concentration of 95%.

ABSTRAK

Staphylococcus aureus merupakan bakteri patogen yang dapat ditemukan pada ulkus diabetik dan berisiko menghambat proses penyembuhan luka. Penggunaan antibiotik yang tidak rasional dapat meningkatkan risiko resistensi bakteri sehingga diperlukan eksplorasi bahan alam sebagai alternatif antibakteri. Penelitian ini bertujuan menganalisis aktivitas antibakteri infusa bawang putih (Allium sativum) terhadap S. aureus isolat klinis dari ulkus diabetik berdasarkan jumlah koloni bakteri. Infusa dibuat dalam konsentrasi 50%, 65%, 80%, dan 95%, dengan empat kali pengulangan pada setiap kelompok. Pengujian dilakukan menggunakan metode dilusi cair yang dilanjutkan dengan penanaman pada media padat dan penghitungan koloni. Data dianalisis menggunakan uji Shapiro-Wilk, Levene, dan One-Way ANOVA pada taraf signifikansi 5%. Hasil penelitian menunjukkan terdapat perbedaan bermakna jumlah koloni antar kelompok konsentrasi (p < 0,05), dengan jumlah koloni yang semakin rendah seiring peningkatan konsentrasi infusa dan penghambatan tertinggi ditemukan pada konsentrasi 95%. Temuan ini menunjukkan bahwa infusa bawang putih memiliki aktivitas antibakteri terhadap S. aureus isolat klinis ulkus diabetik dan menunjukkan potensi sebagai kandidat antibakteri berbasis bahan alam, khususnya pada konsentrasi 95%.
